<scp>ThPOK</scp> inhibits osteoclast formation via <scp>NFATc1</scp> transcription and function

Both LRF (Zbtb7a) and ThPOK (Zbtb7b) belong to the POK (BTB/POZ Kruppel) family of transcription repressors which participate in development, differentiation oncogenesis. While mediates osteoclast by regulating NFATc1 expression, principal established function is transcriptional control T-cell lineage commitment. Whether affects formation or function, not known.We find that marrow macrophage expression diminishes with exposure RANKL but deficiency does affect differentiation. On other hand, enhanced ThPOK, macrophages, substantially impairs osteoclastogenesis. Excess binds promoter suppresses its suggesting a mechanism for inhibitory effect. Despite suppression osteoclastogenesis excess being associated diminished NFATc1, rescued overexpression. Thus, appears inhibit osteoclastogenic capacity while depletion has no effect on bone resorptive cell. This article protected copyright. All rights reserved.
